aGVHD對小鼠腸道血管形態與結構的影響及其機制

摘要:為了探究aGVHD發病進程對小鼠腸道組織血管的變化和腸道免疫穩態之間的關系和機制。我們建立小鼠aGVHD模型,分析小鼠小腸絨毛血管內皮細胞和周細胞以及浸潤免疫細胞在不同時間點的動態變化,并運用qPCR、Smart-seq技術分析尋找差異表達基因。發現在aGVHD的發病進程中,小腸絨毛血管的損傷及浸潤免疫細胞的變化與疾病進程密切相關,與炎癥反應、細胞凋亡及腸道屏障相關基因表達顯著變化。根據研究結果,我們認為aGVHD誘發的小腸絨毛血管的嚴重損傷和結構紊亂可能是導致受體死亡的一個重要原因,小腸絨毛的aGVHD炎癥反應可能主要通過影響與內皮細胞凋亡及腸道屏障相關的基因表達,從而破壞小腸絨毛血管的完整性和提高其通透性。

The effects of aGVHD on the morphology and structure of murine intestinal blood vessels and its underlying mechanism
Abstract:To explore the relationship and mechanism between the changes of intestinal tissue blood vessels and intestinal immune homeostasis in mice during the course of aGVHD. We established a mouse aGVHD model, and analyzed the dynamic changes of endothelial cells, pericyte and infiltrating immune cells in mouse small intestinal villi at different time points. qPCR and Smart-seq were used to search for differentially expressed genes. It was found that in the course of aGVHD, the damage of small intestinal villus vessels and the changes of infiltrating immune cells were closely related to the course of the disease, and the expression of inflammatory response, apoptosis and intestinal barrier related genes were significantly changed. According to the results of the study, we believe that the severe damage and structural disorder of villi vessels induced by aGVHD may be an important cause of receptor death, and the aGVHD inflammatory response of villi may damage the integrity of villi vessels and improve their permeability mainly by affecting gene expression related to endothelial cell apoptosis and intestinal barrier.
